O episódio piloto de "Mister Ed" foi gravado em 1958, sob o título "The Wonderful World of Wilbur Pope". Ninguém sabe ao certo o motivo do mesmo nunca ter ido ao ar. Em Janeiro de 1961 a série propriamente dita estreou através de um sistema chamado Syndication (exibição via canais regionais locais americanos). Foi a primeira série americana a ser lançada no meio de uma temporada (anos depois, em Janeiro de 1966, "Batman" repetiria tal feito). A partir da segunda temporada, "Mister Ed" passou a ser exibida pela Rede CBS.

A estrela do programa era um cavalo chamado Mister Ed, da raça palomino, que podia falar. Seu dono era o atrapalhado arquiteto Wilbur Post (Alan Young). O humor de cada episódio devia-se ao fato de que o cavalo só falava com seu dono, além de ser chegado a uma boa encrenca.

Wilbur tinha uma esposa charmosa e tolerante chamada Carol (Connie Hines). Seus vizinhos eram os Addisons (até 1963, ano da morte de Larry Keating) e os Kirkwoods. Na última temporada, o programa ficou centrado na vida doméstica dos Posts. Após o final da série, Connie Hines deixou o meio artístico mas continuou a fazer aparições públicas ao lado de Alan Young.

O truque utilizado para que o cavalo mexesse a boca dando a impressão de falar foi por muitos anos um mistério. Existe uma versão da qual contam que pasta de amendoim era utilizada para que ele movimentasse a boca sem parar. No ano de 2004 o ator Alan Young teria admitido que essa história foi inventada por ele e que na verdade era colocada na boca de Ed um pedaço de nylon, que o cavalo então tentava remover. Existem versões não confirmadas que cabos finos é que faziam a cabeça do cavalo se movimentar.

A série derivou-se de um conto chamado "Ed Takes The Pledge", de Walter R Brooks, da série infantil "Freddy, The Pig". As histórias eram baseadas em animais que conversavam e interagiam com os humanos. Anos depois, o ator Eddie Murphy estrelou um filme chamado "Dr. Dolittle" onde algumas pessoas aparecem assistindo "Mister Ed" e falando sobre o mito da pasta de amendoim.

"Mister Ed" trouxe vários astros convidados, dentre eles, Mae West, Clint Eastwood, Zsa Zsa Gabor, Donna Douglas e Alan Hale Jr. A fábrica de automóveis Studebaker patrocinou o programa de 1961 a 1963. Daí até 1965 a série foi patrocinada pela Ford Motor Company.

Em 2004 foi planejado um remake pela fox, com Sherman Hemsley fazendo a voz de Mister Ed, David Alan Basche como Wilbur e Sherilyn Fenn como Carol. O piloto foi filmado mas não chegou a ser exibido. O lançamento da série em DVD ocorreu nos Estados Unidos entre 2009 e 2011. Em 2012, a Waterman Entertainment anunciou estar desenvolvendo um novo projeto envolvendo a produção de um longa metragem.

No Brasil, "Mister Ed" estreou pela TV Bandeirantes Canal 13 de São Paulo, no sábado de 20 de Maio de 1967, as 18h15. Após seu término de exibição na emissora, migrou para a TV Record Canal 7 de SP e na sequência para a TV Tupi Canal 4 de SP. Sua última exibição entre nós ocorreu através do canal MGM, da TV a Cabo.
